Purine-37 Is A Platform For The First Codon/Anticodon Base Pair on

tRNALysdecoding of the codon AAA requires the modification N6-threonylcarbamoyl-adenosine-37 (t6A37) 3'-adjacent to the anticodon. The modified nucleoside is a hydrophobic interaction and stabilization platform for the first anticodon-codon base pair. Here, we present the 3Å resolution structure of the ribosome 30S subunit with the t6A37-modified tRNALys anticodon stem and loop domain bound to its cognate codon in the ribosome's A-site. The presentation is a 'movie', in "QuickTime", allowing you to sense the dimensions of the ribosome, anticodon stem and loop and mRNA codon, as well as the critical positioning of the t6A37. The ribosomal RNA (rRNA) is seen in gray with the ribosomal proteins in tan. The anticodon stem and loop is in yellow, the t6A37 in red, and the mRNA AAA codon in blue. As we zoom in on the decoding site, the helix 44 of rRNA is in green. As we get closer the only helix 44 rRNA residues left are three nucleosides in dark green. These rRNA residues recognize the correctness of the anticodon-codon architecture. Two, A1493 and A1492, fold into the minor grove created by the anticodon-codon interaction and H-bond to the tRNA and mRNA backbones near the first and second base pairings, respectively. In addition, the conformation of G530 goes from cis to anti and it H-bonds to stabilize the third base pair. The movie ends at a crystal orientation highlighting the position of t6A37 as platform for the first base pair. |
